The Apparatus of HPLC
In HPLC, the pump is responsible for delivering the mobile phase at a controlled flow rate (a). However, air bubbles can easily dissolve in the mobile phase under standard atmospheric pressure, which can cause issues like flow rate fluctuations and baseline noise/drift. To prevent this, a degassing unit is used to remove air bubbles from the mobile phase (b). Once the air bubbles have been removed, the mobile phase is delivered to the column. The sample injector then introduces a standard or sample solution into the mobile phase (c). To ensure consistent separation of compounds in the column, a column oven is used to maintain a constant temperature (d). As compounds elute from the column, they are detected by a detector placed downstream of the column (e). The detector signal is processed by a workstation to obtain a chromatogram, which is used to identify and quantify the individual compounds (f).
